Primary reaction: Allergens bind to antibodies solidified on the plate. |

Secondary reaction: The enzyme-labeled antibody binds to the allergens bound to the antibody on the plate in a sandwich-like fashion. |

Enzymatic reaction: When an enzyme substrate is added, color is produced by enzymatic decomposition. The degree of this coloration is measured. |
